چکیده

A phagocyte-type NADPH oxidase complex is a major source of endothelial reactive oxygen species (ROS) production, but its biochemical function and regulation remain unclear. In neutrophils, the p47 subunit is centrally involved in oxidase activation in response to agonists such as phorbol-12-myristate-13-acetate (PMA). We investigated the role of p47 in endothelial cell ROS production in response to PMA or tumor necrosis factor(TNF ) stimulation. To specifically address the role of p47, we studied coronary microvascular endothelial cells (CMECs) isolated from p47 / mice and wild-type controls. p47 was absent in hearts of knockout mice whereas the essential oxidase subunit, p22, was expressed in both groups. In the absence of agonist stimulation, the lack of p47 did not result in a reduction in NADPH-dependent ROS production in p47 / CMECs compared with wild-type CMECs. Prestimulation with PMA (100 ng/mL) or TNF (100 U/mL) for 10 minutes significantly increased NADPH-dependent O2 production in wild-type CMECs, assessed either by lucigenin (5 mol/L) chemiluminescence or dichlorohydrofluorescein (DCF) fluorescence. This response was completely lost in p47 / cells. Transfection of the full-length p47 cDNA into p47 / CMECs caused expression of p47 protein and restoration of the O2 response to PMA and TNF . In wild-type CMECs, transfection of antisense p47 cDNA substantially reduced p47 expression and caused loss of the O2 response to PMA and TNF . These data show that endothelial cell p47 is critical in the upregulation of NADPH oxidase activity by PMA and TNF . (Circ Res. 2002;90:143-150.)

Aldosterone Induces Oxidative Stress Via NADPH Oxidase and Downregulates the Endothelial NO Synthesase in Human Endothelial Cells

Aldosterone is traditionally viewed as a hormone regulating electrolyte and blood pressure homeostasis. Recent studies suggest that Aldo can cause microvascular damage, oxidative stress and endothelial dysfunction. However, its exact cellular mechanisms remain obscure. This study was undertaken to examine the effect of Aldo on superoxide production in human umbilical artery endothelial cel...

17h-Estradiol inhibits NADPH oxidase activity through the regulation of p47 mRNA and protein expression in THP-1 cells

In this report, we demonstrate that NADPH oxidase is activated by tumor necrosis factor-a (TNF-a) plus interferon-g (IFN-g) in human monocytic cells (THP-1 cells) differentiated with phorbol ester (PMA) and that physiological concentration of 17h-estradiol inhibits NADPH oxidase activity in THP-1 cells stimulated with TNF-a plus IFN-g. This effect is mediated by estrogen receptor based on estro...

Diabetes induces pulmonary artery endothelial dysfunction by NADPH oxidase induction.

Recent data suggest that diabetes is a risk factor for pulmonary hypertension. The aim of the present study was to analyze whether diabetes induces endothelial dysfunction in pulmonary arteries and the mechanisms involved. Male Sprague-Dawley rats were randomly divided into a control (saline) and a diabetic group (70 mg/kg(-1) streptozotocin). After 6 wk, intrapulmonary arteries were mounted fo...

The phosphoinositide-binding protein p40phox activates the NADPH oxidase during FcγIIA receptor–induced phagocytosis

Superoxide produced by the phagocyte reduced nicotinamide adenine dinucleotide phosphate (NADPH) oxidase is essential for host defense. Enzyme activation requires translocation of p67(phox), p47(phox), and Rac-GTP to flavocytochrome b558 in phagocyte membranes. To examine the regulation of phagocytosis-induced superoxide production, flavocytochrome b558, p47(phox), p67(phox), and the FcgammaIIA...
